چکیده
Two chaperones, Atp23p and Atp10p, were previously shown to regulate the assembly of yeast mitochondrial ATP synthase, extra expression ATP23 was found partially rescue an atp10 deletion mutant, by unknown mechanism. Here, we identified that residues 112-115 (LRDK) required for its function in assisting demonstrated both functions Atp23p, processing subunit 6 precursor partial mutant. By chasing labeling with isotope 35S-methionine, stability synthase increased null strain upon overexpression ATP23. Further co-immunoprecipitation (Co-IP) blue native PAGE experiments showed Atp10p physically associated each other wild type. Moreover, revealed level mutant compared Furthermore, that, after 72 hours growth, leaky growth on respiratory substrates, presence low recovery oligomycin sensitivity ATPase activity. characterization 24 These results indicated, ATP10 deficiency could be complemented stabilizing some synthase. Taken together, this study two chaperones coordinated which advanced our understanding mechanism

Modular assembly of yeast mitochondrial ATP synthase.
The mitochondrial ATP synthase (F(1)-F(0) complex) of Saccharomces cerevisiae is a composite of different structural and functional units that jointly couple ATP synthesis and hydrolysis to proton transfer across the inner membrane. In organello, pulse labelling and pulse-chase experiments have enabled us to track the mitochondrially encoded Atp6p, Atp8p and Atp9p subunits of F(0) and to identi...

The Atp9p ring is one of several assembly modules of yeast mitochondrial ATP synthase. The ring, composed of 10 copies of Atp9p, is part of the rotor that couples proton translocation to synthesis or hydrolysis of ATP. We present evidence that before its assembly with other ATP synthase modules, most of Atp9p is present in at least three complexes with masses of 200-400 kDa that co-immunopurify...

We used electron cryotomography of mitochondrial membranes from wild-type and mutant Saccharomyces cerevisiae to investigate the structure and organization of ATP synthase dimers in situ. Subtomogram averaging of the dimers to 3.7 nm resolution revealed a V-shaped structure of twofold symmetry, with an angle of 86° between monomers. The central and peripheral stalks are well resolved. The monom...
متن کاملErratum to: The Mitochondrial Permeability Transition Pore and ATP Synthase.
Mitochondrial ATP generation by oxidative phosphorylation combines the stepwise oxidation by the electron transport chain (ETC) of the reducing equivalents NADH and FADH2 with the generation of ATP by the ATP synthase. Recent studies show that the ATP synthase is not only essential for the generation of ATP but may also contribute to the formation of the mitochondrial permeability transition po...
متن کاملThe INA complex facilitates assembly of the peripheral stalk of the mitochondrial F1Fo-ATP synthase.
Mitochondrial F1Fo-ATP synthase generates the bulk of cellular ATP. This molecular machine assembles from nuclear- and mitochondria-encoded subunits. Whereas chaperones for formation of the matrix-exposed hexameric F1-ATPase core domain have been identified, insight into how the nuclear-encoded F1-domain assembles with the membrane-embedded Fo-region is lacking. Here we identified the INA compl...
